XL114 mechanism of action (MOA) animation. XL114 is a small molecule inhibitor of the CARD11-BCL-MALT1 (CMB) complex, a key regulator of canonical NF-kappaB signaling downstream of B cell and T cell receptor activation. The precise MOA of XL114 is not known. © 2022 Exelixis, Inc.

The project
This innovative mechanism-of-action (MOA)animation was created to educate HCPs on the role of CARD11-BCL10-MALT1 (CBM) in cancer and introduce the MOA of CBM inhibitor, XL114. We designed an animated schematic to explain how CBM bridges B-cell and T-cell receptor signaling to the NF-κB pathway. Components were modeled using protein structure data and mapped according to subcellular localization data. Multiple data were used to animate highly-complex CBM filament assembly.
